The England international centre-back missed Sunday's home win against Aston Villa with a groin injury and he will not have recovered in time for the quarter-final 1st leg tie at Old Trafford on Tuesday night.
Brazilian midfielder Anderson is also unlikely to have recovered from the thigh injury that he sustained whilst on international duty last week.
Manager Sir Alex Ferguson will, however, have Nemanja Vidic, Paul Scholes and Wayne Rooney available for selection for the Porto game - all three were suspended for the league game against Villa on Sunday.
"I don't think any of the injured players will be back against Porto," Fergie told MUTV.
"But we do have the suspended players back which is good. Vida will play alongside Jonny Evans at the back with John O'Shea at right-back.
"Rooney and Scholes are also available which will give us a bit more penetration and experience.
"I'll have to assess how Ryan Giggs is after playing the 90 minutes against Villa. It's a big ask for someone at 35 years of age to play another game two days later at the kind of level we play."
