The United and England international defender has an ankle injury which kept him out of the side that won against West Ham in midweek and he is unlikely to have recovered in time for the game against The Tigers on Saturday.
Michael Carrick, however, could return to the starting line-up having been named amongst the substitutes for the Hammers match.
"In my programme notes, I've said that I can't believe Hull are above United in the table, and I don't mean that to sound derogatory," Fergie has said of the high-flying Tigers.
"I've always said when you come out of the Championship you need to invest to survive in the Premier League.
"Hull have gone about it a completely different way. We can't treat the game lightly.
"We took our foot off the pedal too much in the second half against West Ham and we can't afford to do that against Hull."
